The official website of the University of Tokyo in Japan recently announced that researchers at the University of Tokyo and Kyoto University have discovered the molecular mechanism of RNA interference (RNAi). The so-called ribonucleic acid interference is a phenomenon in which certain protein synthesis occurs when single-molecule RNA is split.

Since RNAi can shut down the expression of specific genes, scientists have been expecting RNAi to be used in the medical field. In previous studies, scientists have discovered that RNAi is regulated by the RNA-induced silencing complex (RISC), and that the center of the RISC contains a small RNA and an alpha protein that can cleave the target RNA. However, the scientific community has not found a tool to directly monitor RNAi responses, so the molecular mechanism of RNAi has also been a mystery.

For the first time, Japanese researchers have designed a single-molecule imaging analysis method that can observe the process of RISC splitting target RNA in real time in a test tube. They observed that the small RNA in RISC contains two parts, some of which are quickly "bound" to the target RNA to be split, and the other part is responsible for "calibration" whether the correct target RNA is selected.

This breakthrough result reveals the mechanism of action of RISC, or will accelerate the scientific research on RNAi, such as the development of RNA-based gene therapy drugs that inhibit the growth of certain pathogenic proteins.
